Vitamin D is a steroid Vitamin, a group of fat-soluble prohormones that help maintain the balance of calcium in the body, important for normal growth and mineralization of bones and teeth. Extensive and ongoing research on Vitamin D has also brought to light new roles of this Vitamin in modulation of cell growth, neuromuscular and immune functions.

Vitamin D - Background

In the 1920's Edward Mellanby and Elmer McCollum discovered an anti-rachtic factor in cod liver oil while researching a cure for the disease rickets. McCollum identified the factor to be distinct

from Vitamin A in 1922 and called it Vitamin D as it was the fourth Vitamin to be named.

In 1927 Dr. E. H. Reerink and Dr. A. van Wijk, two young scientists in the Phillips’ Physics Laboratory in Eindhoven, the Netherlands, used the newly developed high pressure ultraviolet lamp to obtain reaction products from ergosterol by careful wavelength monitoring. They achieved the first industrial scale photochemical transformation of ergosterol into an anti-rachtic substance which was a synthetic method of producing pure Vitamin D. Vitamin D3 – Synthesis & metabolism in the body


The epidermis and dermis layers of the skin contain 7-dehydrocholesterol also known as provitamin D3 which on exposure to Ultra Violet radiation between 290 – 315 nm (UVB range) is synthesized to Vitamin D3.

Vitamin D3 synthesized on the skin and consumed in food is converted into the metabolite 25(OH)D3 i.e. 25-hydroxyvitamin D3 or calcidiol in the liver in the presence of the enzyme 25-hydroxylase.

The metabolite 25(OH)D3 is the major circulating form of Vitamin D3 in the blood.

In the kidney 25(OH)D3 is converted into 1,25(OH)2D3 i.e. 1,25 dihydroxyvitamin D3 or calcitriol which is the active form of Vitamin D3 required by the body.

1,25(OH)2D3 regulates calcium metabolism through its interaction with its major target tissues, the bone and the intestine. Various factors, including serum phosphorus (Pi) and parathyroid hormone (PTH), regulate the renal production of 1,25(OH)2D3.
